ABSTRACT

OBJECTIVE To establis h the fingerprint of Cynanchum auriculatum,to conduct its chemical pattern recognition analysis,and to determine the contents of four components at the same time. METHODS High performance liquid chromatography (HPLC)method was adopted. The determination was performed on ACE UExcel C 18 column with mobile phase consisted of acetonitrile-0.1% phosphoric acid solution (gradient elution )at the flow rate of 1.2 mL/min. The determination wavelength was set at 210 nm,and the column temperature was 40 ℃. The sample size wa s 10 μL. Taking qingyang shengenin as the reference ,HPLC fingerprints of 16 batches of C. auriculatum medicinal materials were drawn and similarity was evaluated by using the Similarity Evaluation of Chromatographic Fingerprints of Traditional Chinese Medicine (2012 edition), and the common peaks were determined. SPSS 26.0 software andSIMCA 14.0 software were used for cluster analysis ,principal component analysis and orthogonal partial least squares- discriminant analysis. The differential components affecting the quality of C. auriculatum were screened by taking the value of variable importance in projection (VIP)greater than 1 as the standard ;same HPLC method was used to determine the contents of syringic acid ,acyl asclepiadelenin ,baishouwubenzophenone and qingyang shengenin. RESULTS There were 29 common peaks in 16 batches of C. auriculatum ,with a similarity of 0.723-0.998. Four common peaks were identified ,namely syringic acid (peak 7),acyl asclepiadoidin (peak 9),baishouwubenzophenone(peak 13)and qingyang shengenin(peak 15). The results of cluster analysis showed that 16 batches of C. auriculatum could be clustered into three categories ,among which S 1 were grouped into one category,S3 were grouped into one c ategory,S2,and S 4-S16 were grouped into one category. The results of principal component analysis showed that the cumulative variance contribution rate of the five principal components was 88.706%,and the classification results were consistent with the results of cluster analysis. The results of orthogonal partial least squares-discriminant analysis showed that the common peaks (from large to small )with VIP value greater than 1 were peak 20,peak 10,peak 25,peak 12, peak 15(qingyangshengenin),peak 21,peak 14,peak 16,peak 26,peak 22 and peak 17. The linear ranges of syringic acid,acyl asclepterin,baishouwubenzophenone and qingyangshengenin were 0.715 3-45.778 0,2.379 4-152.281 0,0.642 0- 41.085 0,14.541 6- 930.662 0 µg/mL respectively (all R 2>0.999). The quantitative limits were 0.357 7,0.475 9,0.642 0 and 2.423 6 μg/mL;the detective limits were 0.146 0,0.164 1,0.248 8 and 0.833 3 μg/mL,respectively. RSDs of precision ,repeatability and stability (24 h)tests were less than 3%;the average recoveries were 99.11%(RSD=2.00%,n=9),98.54%(RSD=2.21%,n=9), 96.33%(RSD=2.54%,n=9)and 95.96%(RSD=2.93%,n=9);the contents were 17.12-147.80,95.23-583.10(S8 below the quantitative limit ),16.91-210.88 and 211.68-3 587.15(S1 below the quantitative limit )μg/g,respectively. CONCLUSIONS Established HPLC fingerprint and the method of content determination are stable ,reliable,accurate and reproducible. Combined with analysis of chemical pattern recognition ,it can be used for the quality control of C. auriculatum .

ABSTRACT

To explore the optimal therapy time for the treatment of severe coronavirus disease 2019(COVID-19)by traditional Chinese medicine(TCM)and its influence on the therapeutic effect and prognosis. The clinical data,laboratory findings,and outcomes of 64 patients with severe COVID-19 treated with TCM and western medicine in Chongqing from January 20,2020, to March 11,2020 were retrospectively analyzed.Patients were divided into early intervention group[TCM was initiated within 3 days (including day 3) after the first diagnosis of severe type/critical type COVID-19]and late intervention group[TCM was initiated after 7 days (including day 7) after the first diagnosis of severe type /critical type COVID-19].The changes in clinical parameters during the course of disease were compared between the two groups. On day 14,the oxygenation index was 292.5(252.0,351.0)mmHg in the early intervention group,which was significantly higher than that in the late intervention group [246.0(170.0,292.5)mmHg](=0.005).The length of hospital stay [(18.56±1.11)d (24.87±1.64)d,=0.001],duration of ICU stay [(14.12±0.91)d (20.00±1.53)d,=0.000] and time to negativity [(16.77±1.04)d (22.48±1.66)d,=0.001] in the early intervention group were significantly shorter than those in the late intervention group.The intubation rate(7.3%)in the early intervention group was significantly lower than that in the late intervention group(30.4%)(=0.028). Early TCM therapy within three days after a diagnosis of severe COVID-19 can shorten the length of hospital stay,duration of ICU stay,and time to negativity and decrease intubation rate.

ABSTRACT

Objective To analyze the death and disease burden of injury in Guangxi from 2014 to 2016, so as to provide scientific basis for making policy strategies on injury prevention. Methods Datas from the Death Monitoring Network in Guangxi from 2014 to 2016 were analyzed. Descriptive statistics and Chi square were used to describe the mortalities. The potential years of life lost (YPLL) and average years of potential life lost(AYPLL) were assessed. Results A total of 29 288 deaths were reported in Guangxi. The mortality rate was 42.77/100 000. The overall mortality rate decreased over the past three years. Traffic accident, accidental fall, drowning, suicide and sequela, accidental poisoning were the top five causes of injury deaths. The leading cause of injury death in the 0 to 14 age group was drowning, traffic accident in the 15 to 64 age group and accidental fall in the 65 years old and above group. The AYPLL was 29.11 years and, which was higher in male than in female and there was no significant difference between urban and rural. Suicide and sequela had become the biggest average disease burden. Conclusions The average mortality rate decreased in Guangxi from 2014 to 2016. It is necessary to take effective measures to prevent and control injury.

ABSTRACT

Guominjian is a prescription created by famous traditional Chinese medicine (TCM) doctor ZHU Zhan-yu. The prescription is composed of four TCM:Saposhnikoviae Radix, Mume Fructus, Schisandrae Chinensis Fructus and Stellariae Radix, which are commonly used in clinical practice for allergic skin diseases, such as urticaria, atopic dermatitis and eczema. Although the medicinal composition is simple, the curative effect is remarkable, and the total effective rate of treatment in clinic is as high as 90%. Guominjian has the effects on phlegm and blood stasis, and can relieve itching and reduce capillary permeability by antagonizing histamine, reducing serum immune globulin (Ig) E, inhibiting mast cell degranulation and changing the differentiation of CD4+ T cell subsets, with anti-allergy effect at multi-target, multi-path, multi-levels. Domestic and foreign scholars have screened out the chemical constituents of single herbs in Guominjian, and found anti-allergic active ingredients, such as α-cubebenoate, cimicin, schisandrin, and saposhnikovia divaricata polysaccharide, and the mechanisms are different. However, the effects of TCM compounds are not equal to the simple combination of single herbs, and it is not possible to measure the efficacy of an active ingredient in a single herb. The anti-allergic chemical composition in Guominjian should be comprehensively analyzed to study the anti-allergy effect of Guominjian in an all-round way. However, the current research on Guominjian is only limited to the pharmacodynamics, but there is still lack systematic, comprehensive and holistic understanding of anti-allergic active components in Guominjian, which limits the secondary development and the establishment of standards of Guominjian. Therefore, the authors systematically collect and summarize the anti-allergic modern research results of Guominjian and its components by searching domestic and foreign literatures, in order to further define the research and new mechanism of anti-allergic chemical components and mechanism of Guominjian, and provide reference for the development of anti-allergic new drugs.

ABSTRACT

Objective: Cimicifuga dahurica (Ranunculaceae) has many bioactivities. Although there have been intensive studies for saponin constituents at present, only a few studies have focused on for chemical constituents of phenolic acid. To define the phenolic acid constituents,C. dahurica was separated, and the structures of the compounds were identified,in the expectation of providing a basis for its further development,utilization and quality control. Method: A total of 16 kg rhizome of C. dahurica was extracted with 70%ethanol for three times by heating reflux. These 3 extracts were decompressed and concentrated,and then dissolved in water. Then the solvent was successively extracted with petroleum ether,ethyl acetate(EtOAc) and n-butanol(BuOH). The components of EtOAc and water extract were isolated and purified by macroporous,silica gel,ODS,Sephadex LH-20 column chromatography,preparative HPLC and recrystallization,and the structures were identified by nuclear magnetic resonance(NMR) and physicochemical analysis etc. Result: Fifteen compounds were isolated from the ethyl acetate and water fractions,and identified as cimicifugic G (1),2-caffeoyl piscidic acid (2),cimicifugic A (3),cimicifugic B (4),caffeic acid 3-O-β-D-glucopyranoside (5),cimicifugic E (6),cimicifugic F (7),trans-ferulic acid 4-O-β-D-glucopyranoside (8),carboxymethyl isoferulate (9),3,4-dimethoxycinnamic acid (10),ethyl ferulate (11),caffeic ester glucoside (12),shomaside A (13),isoferulic acid (14),caffeic acid (15). Conclusion: Compounds 1-7,9-10,13 were isolated from the plant for the first time.

ABSTRACT

OBJECTIVE: To observe the relationship between the analgesic effect of balance acupuncture and functional changes in brain in patients with migraine without aura. METHODS: A total of 40 cases of migraine without aura were equally randomized into a headache-acupoint group and a sham-acupoint group. When acupuncture given, a filiform needle was inserted into the headache-acupoint (the midpoint of the depression region anterior to the juncture of the first and second metatarsal bones on the dorsum of the foot) or the sham point (the midpoint of the depression region anterior to the juncture site between the 3rd and 4th metatarsal joints of the dorsum of the foot) about 25-40 mm deep and manipulated for a while till the patient experienced feelings of electric shock and numbness, then withdrawn immediately. The treatment was conducted once daily for 4 weeks. The visual analogue scale (VAS) was used to evaluate the severity of pain, and the regional homogeneity (ReHo) analysis of resting state functional magnetic resonance imaging (fMRI) was used to assess changes of the spontaneous brain activity. RESULTS: After acupuncture, the analgesic effect of headache-acupoint was better than that of the sham-acupoint in both intervention stage and the follow-up stage (P0.05). Compared with pre-intervention, 4-weeks' intervention at the headache-acupoint showed an increase of ReHo values in the anterior cingulate gyrus, anterior central gyrus, superior orbital frontal gyrus, insula, inferior lobule, left anterior cingulate gyrus, ventral lateral nucleus and ventral posteromedial nucleus of the thalamus, pontine nucleus, cerebellar tonsils and orbital frontal inferior gyrus of the brain (P<0.05), and a decrease of ReHo values in the right brain bridge, central posterior gyrus, posterior cingulate gyrus, left central anterior gyrus, posterolateral nucleus of thalamus, and hippocampus (P<0.05), separately. In the sham-acupoint group, the ReHo value was increased in the right tongue gyrus, the left anterior lobe, the anterior cingulate gyrus and the lower occipital gyrus of the brain (P<0.05), and reduced in the left ventral posterolateral nucleus of the thalamus, separately (P<0.05). CONCLUSION: Balance acupuncture stimulation of headache acupoint has an analgesic effect in migraine patients without aura, which may be related to its effect in regulating resting state brain function of the limbic-system-dominated multiple brain regions.

ABSTRACT

Objective To investigate the cytoprotection and mechanism of carbachol(CCH)to stimulate M3mus-carinic acetylcholine receptor(M3-mAChR) against hypoxia injury induced by cobaltous chloride hexahydrate (CoCl2) in rat cardiomyocyte line H9c2. Methods Select the normal rat cardiomyocyte line H9c2 as the control group, rat cardiomyocyte line H9c2 was managed with CoCl2to develop hypoxia injury model, M3-mAChR spe-cific agonist CCH and antagonist 4-diphenyl-acetoxy-N-methyl-piperidine methiodide(4-DAMP) were used for in-tervention. The cell viability was tested by 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide (MTT); The apoptosis in cardiomyocyte was detected by flow cytomery(FCM); The expression of M3-mAChR,caspase-3, HIF-1α and HO-1 proteins was measured by Western blot assay. Results In hypoxia group,the ap-optosis rate was significantly increased while cell proliferation decreased, the expression of HIF-1α, caspase-3 and HO-1 proteins were up-regulated obviously;After treatment with CCH,the apoptosis and cell proliferation of cardiomyocytes were significantly decreased, while the proliferation of cells increased, and the expression of M3-mAChR, HIF-1α and HO-1 proteins increased, the expression of caspase-3 protein was significantly decreased. Moreover, when applying 4-DAMP as intervention, these effects mediated by CCH was abolished.Conclusions CCH stimulates M3-mAChR against hypoxia injury induced by CoCl2in rat cardiomyocyte strain H9c2, and the mechanism may be related to down-regulation of caspase-3 expression and up-regulation of HIF-1α and HO-1 protein expression.

AIM:To measure ocular biometric values with sexual and age and determine the relationship between the differences using the Lenstar 900.METHODS:Totally 413 myopes 826 eyes,200 males (400 eyes) and 213 females (426 eyes),were enrolled in this study and were divided into 3 groups:Group Ⅰ (< 5 years),Group Ⅱ (5-10 years),Group Ⅲ (>10 years).Central corneal thickness (CCT),anterior chamber depth (ACD),lens thickness (LT),axial length (AL),white-to-white distance (WWD) and pupil diameter (PD) were measured by Lenstar 900.The differences between age groups and gender groups were compared using the LSD and SNKk methods in variance analysis.Pearson correlation coefficient to assess AL,CCT,ACD,LT,WWD,PD in children and adolescents.RESULTS:There were significant difference in CCT between ages groups (P<0.05) which increased with the age.There were significant differences both in ACD and AL between sexual groups.With analysis of Person,CCT showed a significantly positive correlation with WWD and PD(r=0.208,0.167;P<0.05) and ACD showed a significantly positive correlation with AL,WWD,PD(r=0.620,0.238,0.192;P<0.05).LT showed a significantly negative correlation with ACD,AL and WWD (r=-0.271,-0.186,-0.227;P<0.05).WWD showed a significantly positive correlation with PD (r=0.273,P<0.05).CONCLUSION:CCT has gradually thickening trend with ages.Men are more than women in ACD and AL.CCT shows positive correlation with WWD and PD and ACD shows positive correlation with AL,WND,PD.LT shows negative correlation with ACD,AL and WWD.WWD showed positive correlation with PD.

Objective:To analyze the advantages of the pressure-regulated volume control (PRVC) mode compared with the traditional continuous mandatory ventilation (CMV) mode about mechanical ventilation efficiency of ventilator.Methods: For the mechanical ventilation situations, the parameters of PRVC mode, V-CMV mode and P-CMV mode under same detecting conditions were tested by using gas analyzer, which based on the ventilation principle of PRVC mode. And then the differences between PRVC mode and CMV modes were analyzed. Results: PRVC mode can supply the almost same tidal volume, and provide lower pressure and higher compliance compare with the V–CMV mode under the same conditions. On the other hand, PRVC mode can supply more tidal volume and slightly higher compliance compare with the P–CMV mode under the same pressure.Conclusion: PRVC mode has combined with the advantages of CMV mode, and it can make sure the target tidal volume, at the same time, reduce the pressure of airway, and improve the compliance for circuit. It also can make the mechanical ventilation to got more efficiency and more safety.

Objective To evaluate the serum Prostaglandin E2 (PGE2) level in Acute-on-chronic liver failure (ACLF) and determine its predicative value for infection. Methods From April 2014 to April 2015, ninety-one patients with hepatitis B virus and ACLF but without infection were enrolled into this prospective study that was carried out at our Hospital. Twenty patients with stable chronic hepatitis B were enrolled from the outpatient department and twenty healthy control subjects without any disease were enrolled from hospital staff. Serum PGE2 levels were determined using ELISA at enrollment. Clinical and laboratory parameters were collected. Receiver operating characteristic (ROC) curves were used to determine optimal cut-off values to predict infection. Results Significantly higher PGE2 levels were found in patients with ACLF in comparison with healthy controls and patients with stable CHB (P < 0.000 1). In ACLF patients, PGE2 levels were significantly higher in patients that eventually developed infection than those without this complication (P < 0.000 1). ROC analysis showed that serum PGE2 (area under the ROC curve, 0.83) could predict infection in patients with ACLF with sensitivity of 78.4% and specificity of 81.5% using a threshold of 141 pg/mL. Conclusions Serum PGE2 is associated with the susceptibility to secondary infections for patients with ACLF. Increased PGE2 serum levels may serve as a potential biomarker for developing infections in ACLF patients.

ABSTRACT

Objective To observe the effects of balance acupuncture at hypoglycemic point on type 2 diabetes mellitus.Methods Three-week balance acupuncture on hypoglycemic point was executed for 60 cases of confirmed type 2 diabetes mellitus patients,the changes of blood glucose,urine glucose,systolic pressure,diastolic pressure,blood fat,body weight and clinical symptoms were recorded,and statistical analysis was carried out with CHISS software.Results Balance acupuncture on hypoglycemic point decreased significantly the blood glucose,blood pressure,blood fat and etc of the patients (P<0.001),the clinical symptoms were improved obviously after acupuncture such as thirst,bulimia and dieresis (P<0.001),and there was statistical difference between the body weights before and after acupuncture (P<0.05).Conclusion Balance acupuncture contributes to improving the blood glucose,blood pressure and blood fat of type 2 diabetes mellitus and relieves the clinical symptoms,whose mechanism,involving in the idea of brain science,is to fulfill acupuncture through central control.

ABSTRACT

Learning/memory impairment is one of the most serious problems induced by stress, and the underlying mechanisms remain unclear. Opiates and opioid receptors are implicated in multiple physiological functions including learning and memory. However, there is no clear evidence whether the endogenous opioid system is involved in the formation of the stress-induced spatial reference memory impairment. The aim of the present study was to evaluate the role of μ opioid receptor in the stress-induced spatial reference memory impairment by means of Morris water maze (MWM) test in a mouse elevated platform stress model. The mice were trained in the MWM for four trials a session for 4 consecutive days after receiving the elevated platform stress, and intracerebroventricular injection of μ opioid receptor agonist DAMGO, antagonist CTAP or saline. Retention of the spatial training was assessed 24 h after the last training session with a 60-s free-swim probe trial using a new starting position. The results showed that intracerebroventricular injection of μ opioid receptor agonist DAMGO but not antagonist CTAP before MWM training impaired the memory retrieval of mice. Elevated platform stress before MWM training also impaired memory retrieval, which could be reversed by pre-injection of CTAP, and aggravated by DAMGO. These results suggest that endogenous opioid system may play a crucial role in the formation of the stress-induced memory impairment.

ABSTRACT

Objective To evaluated the application value of mean corpuscular volume(MCV) and red blood cell volume distribu-tion width(RDW) in predicting early treatment responses of non-severe aplastic anemia(NSAA) .Methods 101 cases of patients who were newly diagnosed with non-severe aplastic anemia and were treated with Cyclosporine (CsA)combined androgen therapy . Treatment before baseline MCV ,RDW value were measured ,treatment for 3 months ,6 months test routine blood ,reticulocyte indi-cators and treatment results were statistically analyzed and receiver operating characteristic (ROC) curve was used to estimate the sensitivity and specificity of MCV ,RDW in predicting early treatment results .Results The level of MCV and RDW in non-severe aplastic anemia were significantly higher than normal reference .the responded patients had significantly higher pretreatment base-line MCV value than those non-responded .especially at 6 month ,baseline MCV were 105 .10 fl and 98 .30 fl respectively (P=0 .025) .the responded patients had significantly lower pretreatment baseline RDW value than those non-responded .especially at 3 month .baseline RDW were 14 .60% and 16 .60% respectively(P=0 .001) .The cutoff level of MCV (100 fL) and RDW (15 .55% ) for the predicting 3 and 6 month treatment responses were established based on the ROC curve ,with degree of accurancy of MCV was 62 .4% ,61 .4% and RDW 70 .3% ,63 .4% respectively .compared the treatment efficacy acuity MCV ≥100 fL/RDW <15 .55%group was significantly better than MCV < 100 fL/RDW≥15 .55% group .Multivariate analysis showed that pretreatment RDW and absolute reticulocyte value was the early prognostic factor of NSAA treatment effect .84 patients with ARC ≥ 20 × 109/L , through RDW cutoff Layered compare treatment response :RDW<15 .55% group was significantly better than the RDW≥15 .55%group(P=0 .000) .Conclusion MCV can not serve as of a significant predictor of early treatment response in non-severe aplastic a-nemia .RDW can serve as of the bone marrow failure severity indicators and a significant predictor of early treatment response in non-severe aplastic anemia .The joint reticulocyte absolute value parameters ,which can more accurately predict treatment efficacy .

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the effects of CS(2) occupational exposure on thumbprint and intelligence.</p><p><b>METHODS</b>According to the monitoring results of CS(2) concentration, which were got at various workplace in chymic fiber factory, the workers were divided into three groups including the low-concentration group (TWA < 5 mg/m(3)), the high-concentration group (TWA >or= 5 mg/m(3)) and the control group. The method of cluster sampling was used respectively. 139, 129 and 327 people were taken at random from three groups and they were given thumbprint and intelligence examinations through following three methods, including MMPI, SCL-90 and SPM.</p><p><b>RESULTS</b>MMPI test showed that Hy, Pa, Pt, Sc positive rates in the low-concentration group were 7.19%, 2.16%, 26.62% and 10.07%, Hs, D, Hy, Pa, Pt, Sc, Ma, Si positive rates in the high-concentration group were 32.56%, 8.53%, 9.30%, 24.81%, 2.33%, 42.64%, 15.50%, 5.43% and 6.20%. There was a significant difference with the control group (Hs, D, Hy, Pd, Pa, Pt, Sc, Ma, Si was 14.07%, 2.14%, 2.14%, 14.07%, 0.00%, 17.74%, 3.67%, 1.22%, 1.22% respectively). The differences of Hs, Pt positive rates between high-concentration and low-concentration group were significantly. SCL-90 showed that the total positive rate in the low concentration and the high concentration groups was 15.8% and 20.2%. The positive rate of the basic factors were significantly different among the low-concentration group, the high concentration group and the control group while there was no significant difference in the positive rate between the high concentration and low concentration group. SPM test found that the positive rates of the intelligence in the low concentration group and the high concentration group were 35.1% and 35.7% respectively and the control group (15.1%) was significantly different from low-concentration and high concentration group. Moreover, there was no obvious difference between high-concentration and the low-concentration group. The intelligence damage was associated with the length of CS(2) occupational exposure and the correlation coefficient had statistical significance.</p><p><b>CONCLUSION</b>The low-concentration CS(2) occupational exposure could affect personality and intelligence, and the degree of this influence is associated with the length of CS(2) occupational exposure.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the expression of EVEC in ovarian carcinoma and explore its biological significance.</p><p><b>METHODS</b>The expression of EVEC in 22 specimens of normal ovarian tissues and 63 specimens of ovarian cancers was detected by RT-PCR and Western blotting analysis, respectively.</p><p><b>RESULTS</b>RT-PCR showed that the expression level of EVEC in stage I-II ovarian cancer (0.199 ± 0.014) was significantly higher than that in stage III-IV ovarian cancer (0.155 ± 0.015, P < 0.05), and significantly lower than that in normal ovarian tissues (0.415 ± 0.055, P < 0.05). There was no significant difference between the expression levels of EVEC in primary sites and that in corresponding metastatic sites of ovarian cancer (P > 0.05). Furthermore, the results of Western blot also showed that the protein expression level of EVEC in stage I-II ovarian cancer was also significantly lower than that in normal ovarian tissues (0.179 ± 0.026 vs. 0.543 ± 0.032, P < 0.05), and higher than that in stage III-IV ovarian cancer (0.179 ± 0.026 vs. 0.115 ± 0.023, P < 0.05). The EVEC expression level in the epiploic metastasis of stage I-II ovarian cancer was significantly higher than that of stage III-IV ovarian cancer (0.201 ± 0.028 vs. 0.101 ± 0.037, P < 0.05). The expression of EVEC in ovarian carcinoma had no correlation with age, pathologic classification and histological grade (P > 0.05).</p><p><b>CONCLUSIONS</b>EVEC is closely related with carcinoma metastasis. The expression of EVEC in ovarian cancer and its metastatic sites was remarkably decreased. EVEC may play a negative role in the development and metastasis of ovarian cancer and may be a valuable marker in estimation of the prognosis for patients.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To analyze the effectiveness of Chinese medicine and integrated Chinese and Western medicine for influenza A (H1N1) in the fever clinics and its relevant expenditure.</p><p><b>METHODS</b>A prospective survey on the clinical epidemic observation and follow-up was conducted from July 2009 to October 2009 with a self-developed questionnaire whose contents including the clinical data of the confirmed 149 H1N1 cases and their relevant therapeutic expenditure. The patients were assigned to the Chinese medicine group (22 cases treated by Chinese medicine alone) and integrative medicine group (124 cases treated by both Chinese medicine and Western medicine). The data were processed with descriptive analysis, t test and χ (2), and sum-rank test.</p><p><b>RESULTS</b>The proportion of clinical recovery of Chinese medicine group (81.8%) was higher than that of integrative medicine group (54.8%) with statistical significance (P=0.02). The average fever durations in both groups were 3.5 to 4 days, showing no significant difference (P=0.86). In the comparisons of average cost of Chinese herbs, drugs, therapies, and total cost, those of the Chinese medicine group were lower than those in the integrative group (P=0.01, P=0.00, P=0.00, P=0.00).</p><p><b>CONCLUSIONS</b>The H1N1 patients in the fever clinic who received Chinese medicine treatment had a higher clinical recovery proportion than those who received integrated Chinese and Western medicine treatment with lower medical cost. However, due to small sample size of the Chinese medicine group in the study, the conclusion needs further confirmation by studies with large sample size.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To confirm the activity of non structural protein 1 (NSP1) of Rotavirus (RV) as E3 ubiquitin ligase by experiments and to provide some clues for NSP1 on the pathogenic mechanisms and replication of RV.</p><p><b>METHODS</b>The whole gene and RING deleted mutation gene of NSP1 were coloned into pEGFPC1 expression plasmid, and transfected into human embryonic kidney (HEK) 293 FT cells with pBlue-Script-HA-Ubiquitin. The expression of proteins were proved by using con-focal microscope and western blotting. The ubiquination of proteins were detected by co-immunoprecite.</p><p><b>RESULTS</b>The cellular proteins of HEK293FT are ubiquinated by NSP1 protein and NSP1 protein was self-ubiquinated also.</p><p><b>CONCLUSIONS</b>It revealed that RV NSP1 had the activity of E3 ubiquitin ligase and it may play a role on the modulate mechanisms of ubiquination.</p>

ABSTRACT

OBJECTIVE@#To explore the mechanism of paclitaxel on the protein expression of human cervical carcinoma cell line HCE1.@*METHODS@#The total proteins extracted from paclitaxel-treated HCE1 cells were analyzed by 2-dimensional gel electrophoresis (2-DE), and compared with those from untreated HCE1 cells. The differential proteins were identified by mass spectrometry. Western blot was used to determine the differential expression levels of the 2 proteins.@*RESULTS@#At 24 hour after paclitaxel (0.05 mumol/L) treatment, 2-DE images of paclitaxel-treated and paclitaxel-untreated cells were analyzed. Forty-two differential proteins were found. Twenty-one differential proteins among 42 proteins were analyzed by mass spectrometry, among which 15 proteins were identified, including peptidyl-prolylisomerases A (PPIase A),alpha-enolase,keratin 8,heat shock protein 90, eukaryotic translation initiation factor 1A, and so on.@*CONCLUSION@#Fifteen proteins in human cervical carcinoma cells paclitaxel-treated and paclitaxel-untreated are found by proteomic techniques. These proteins may be involved in the proliferation inhibition of human cervical carcinoma cells by paclitaxel.
